活性中间体有机反应中出现的活泼反应中间体。有机反应一般涉及共价键的断裂和形成两个历程,其方式有同时发生协同反应和分阶段进行两种。多数有机反应是分阶段进行的,过程中出现的活泼反应中间体(A和B),再进一步反应生成最终的稳定产物(A-C):一步反应:C+A-B→[C...A...B]→C-A+B阶段反应:A-B→[A...B]→A+BA+C→[A...C]→C-A协同反应是一步完成的,没有反应中间体,所以只经历一个过渡状态(图1)。而阶段反应(图2),从反应物到产物,要经过两个高点和一个低点,这个低点就代表活性中间体。其能量较反应物和产物均高得多,所以很活泼,但是相对于两个高点(过渡状态1和2)能量又较低,因而能短暂存在。中间体的能量势谷(即图中最低点)越低,就越稳定,寿命也越长。迄今已发现多种各具特色的反应活性中间体。主要有:正碳离子(R+)、负碳离子(R-)、自由基(R·)、卡宾(R2C:),还有复合型中间体,如正离子基R抆和负离子基R刔(见离子基)等物种。中间体极其活泼,寿命很短,难以用常规方法加以分离和提纯。鉴于它们的结构特征,可以在特殊条件(低温或捕集、络合等)下使用现代光谱学及其他化学或物理学的手段来鉴别其结构和反应性能。参考书目W.J.le Noble, Highlights of Organic Chemistry,Marcel Dekker,New York,1974.
